I was wondering if the Urusei Yatsura manga could be coloured using the TV series as a guide?

If you own the Manga books, you can coloured them with pencils and pens. xD

I've done that with a few books I have. xD

I was thinking of using computer software to colour downloaded captures of the UY manga, to make it look like the TV show. Does anyone know if this could be done?

Yes, taking the downloaded captures of the manga and using computer software to colour it can be done.

yes, you copy the downloaded manga page into photoshop or adobe and color it with the bucket button

There is an open-source software called GIMP which is similar to Photoshop.  I could tell you several ways to do it in Photoshop, but you'd have to figure it out for yourself with GIMP.

I'm using photoshop but, even with this powerful tool, is not easy at all to give colours to a black & withe story.

So difficult is to give the right shades of color: first of all the backgronds (otherwise it would seems "flat"), then to the characters !!

It's not impossible but it require so much time.

What could I say more?

I've tried gimp and I am unable to use it. I don't have any other means of colouring it so I'm afraid I won't be able to do it.
